چکیده
Abstract Introduction TIPE-2 has been identified as a negative regulator of both innate and adaptive immunity is involved in several inflammatory diseases. However, the role immune suppression dendritic cells (DCs) transduced with not well studied. Methods In this study, DCs were recombinant adenovirus, then cocultured allogeneic CD4+ or CD8 + T cells. The proliferation, cytokine production activation marker levels cell detected. Results data demonstrated that attenuated after treated DCs. Conclusions These results suggested are capable inducing suppression, which provide promising way for therapeutical strategies transplantation autoimmune

Effective antitumor immunity against murine gliomas using dendritic cells transduced with hTERTC27 recombinant adenovirus
hTERTC27, a 27-kDa hTERT C-terminal polypeptide has been demonstrated to cause hTERT-positive HeLa cell apoptosis and inhibits the growth of mouse melanoma. hTERTC27 has been associated with telomere dysfunction, regulation of gene-regulated apoptosis, the cell cycle and activation of natural killer (NK) cells, but its mechanism of action is not fully understood. Here, we report that dendritic ...
متن کاملGeneration of melanoma-specific cytotoxic T lymphocytes by dendritic cells transduced with a MART-1 adenovirus.
Dendritic cells (DC) are potent stimulators of primary T cell responses. In this study, we demonstrate that DC, genetically engineered to express the MART-1/Melan-A (MART-1) tumor-associated Ag, express MART-1 mRNA and protein, correctly process and present the HLA-A2.1-restricted immunodominant MART-1 peptide (MART-1(27-35)), and serve as potent stimulators of MART-1-specific CTL in vitro. A r...
متن کاملRecombinant Adenovirus Infection of Human Dendritic Cells
Recombinant Adenoviruses (rAd) are widely used as gene delivery vectors in gene therapy and vaccination (Hall et al., 2010, Liu, 2010). These replication incompetent vectors have established safety in humans and possess a number of advantages, such as that high viral titres can be produced efficiently. Several different human rAd types are being intensively investigated in clinical trials for t...
متن کاملTGF-Β1 Transduced Mesenchymal Stem Cells Have Profound Modulatory Effects on DCs and T Cells
Background: Mesenchymal stem cells (MSCs) and transforming growth factor-β1 (TGF-β1) molecules are well known for their immunomodulatory properties and their function in tissue regeneration and remodeling. Objectives: To evaluate the interaction of TGF-β1 engineered MSCs with T cells and dendritic cells (DCs) and their modulatory effect on the immune response. Methods: MSCs and DCs were generat...
متن کاملGenetic immunization for the melanoma antigen MART-1/Melan-A using recombinant adenovirus-transduced murine dendritic cells.
Dendritic cells (DCs) are professional antigen-presenting cells that process and present antigenic peptides and are capable of generating potent T-cell immunity. A murine tumor model was developed to evaluate methods of genetic immunization to the human MART-1/Melan-A (MART-1) melanoma antigen. A poorly immunogenic murine fibrosarcoma line (NFSA) was stably transfected with the MART-1 gene. Thi...
